I have a Garmin Nuvi 265wt. How can I best charge it in a car with no cigarette lighter?

You can buy universal 12v charging adapters at pep boys. It's 2 wires, a ground and a power supply. Run the ground to a bolt that is bolted to metal or the battery, and run the hot to either the battery, your fuse panel, or tie it in to a hot wire, like your stereo or dash light.

There might not be an actual "lighter" but there SHOULD be a socket that the lighter gets plugged into. If not, install a socket.

How do I, for the first time, charge the battery in my Garmin nuvi 265WT GPS on my Mac?

Most come with a USB cable to plug into your computer and usually this cable will also charge it. Others have an AC adapter to change while in other cases, these are extra cost options. I would assume whatever the case, that it has a device to power it while in the car, which will also recharge it.
